Caryophyllene
The peppery terpene, and the only one known to bind a cannabinoid receptor directly. What beta-caryophyllene is and why it is unusual.

Beta-caryophyllene is the peppery terpene, and it is structurally unusual enough that it does not behave like the rest of the list.
What it is
Caryophyllene is found in black pepper, cloves, cinnamon, and hops. In cannabis it produces a warm, peppery, faintly woody note — the spice underneath a profile rather than the top note.
Two things make it structurally distinctive. First, it is a sesquiterpene: larger and heavier than myrcene or limonene, with fifteen carbons rather than ten. That makes it less volatile, so it survives heat, time, and storage better than the lighter compounds — one reason it often persists in older material after the citrus notes have faded.
Second, and more interestingly, its structure includes a cyclobutane ring that is rare in nature and gives it a shape most terpenes do not have.
The thing that makes it unusual
Beta-caryophyllene is the only common cannabis terpene known to bind directly to a cannabinoid receptor.
It is a selective agonist at CB2, the receptor found mainly in peripheral tissue and immune cells rather than in the brain. This has been demonstrated in laboratory work and is well enough established that caryophyllene is sometimes described as a dietary cannabinoid.
That is a genuinely notable fact, and it is also frequently over-read. CB2 is not the receptor primarily associated with the psychoactive experience of cannabis — that is CB1, and caryophyllene does not meaningfully bind it. So "caryophyllene acts on a cannabinoid receptor" is true, and "therefore it changes how high you feel" does not follow from it.
What the CB2 activity means for someone consuming a jar of flower containing 0.4% caryophyllene is, honestly, not established. The receptor binding is demonstrated. The consequence at realistic dietary or inhaled concentrations is not.
What you will see on a COA
Caryophyllene is common, and frequently appears in the top three of a profile. Typical flower reports land somewhere in the 0.1% to 0.8% range.
It is particularly associated with the Cookies and Chemdawg families and with diesel-leaning profiles, though as ever the lab report is a better guide than the name — see why the same strain can feel different.
Because it is less volatile than the lighter terpenes, its proportion of a profile tends to rise as a batch ages, even as the absolute figure falls. An old jar can read as more caryophyllene-forward than it was at test, simply because the myrcene and limonene left faster.
Some reports also list alpha-humulene nearby; the two co-occur in cannabis and in hops, and humulene is usually the smaller of the pair.
Tracking it yourself
Caryophyllene is one of the more trackable terpenes: it is common, it is often present in meaningful quantity, and it is stable enough that the reported figure stays roughly honest for longer than most.
Worth noting alongside it:
- The percentage and its rank in the profile.
- The age of the batch, since caryophyllene's share shifts upward over time.
- Whether humulene is present, since the two travel together.
Then log how the session went, across enough sessions and enough different products that any association has to survive some variety before you take it seriously. The receptor biology is interesting. Whether it shows up in your own record is a separate question, and the only way to answer it is to keep one.
